 自组装胶束递送姜黄素 (Curcumin) 可提高非小细胞肺癌 (NSCLC) 的治疗效果

背景:姜黄素(CUR)具有抗肿瘤的效果,可以促进人非小细胞肺癌 A549 的凋亡。然而,姜黄素的临床应用被其水溶性差和生物利用度低所限制,因此,急需开发一个具有较高水溶性和生物相容性的新型姜黄素载药体系用于临床。
材料和方法:用薄膜水化法制作 mPEG-PLA 载姜黄素的胶束CUR/mPEG-PLA胶束,表征其相关参数并评估其抗肿瘤的效果。
结果:CUR/mPEG-PLA 胶束的粒径为 34.9±2.1nm,分散系数为 0.067-0.168,包封率和载药率分别为 90.2±0.78% 和 9.1±0.07%,姜黄素能够持续的从 CUR/mPEG-PLA 胶束中释放出来并且 A549 细胞对姜黄素的摄取明显增加。CUR/mPEG-PLA 胶束可抑制 A549 细胞的增殖,增加其细胞毒性,诱导 G2/M 期阻滞,促进细胞凋亡。CUR/mPEG-PLA 胶束比游离姜黄素更明显抑制 A549 细胞的迁移和侵袭。此外,CUR/mPEG-PLA 胶束抑制人脐静脉内皮细胞的迁移,侵袭和血管生成,这说明其具有抗血管生成的能力。这些抗肿瘤的效果可能与 VEGFMMP-2MMP-9 和 Bcl-2 表达降低以及 Bax 的表达增加有关。
结论:mPEG-PLA 共聚物可作为姜黄素的有效载体,CUR/mPEG-PLA 胶束具有临床治疗非小细胞肺癌的潜力
